You should use the "Use CTS" option, rather than the "Set RTS line" option.

here it works just fine when either "Set RTS line" or "Use CTS" are set. It doesn't when both are unchecked.

It may work, but the connection will be more reliable when you use the correct handshake option.
The "Set xxx Line" option was introduced to power external interfaces.
